"not a defined route"?

It means that there is not a route in /etc/mail/mailertable, since it
probably is not a domain for which you handle e-mail. Since it’s an
unknown route, no call-ahead (milter-ahead) is performed. It’s a
harmless logging of milter-ahead. It has nothing to do with MailScanner.
