If you are using Postfix, you need

Incoming Queue Dir = /var/spool/postfix/hold
Outgoing Queue Dir = /var/spool/postfix/incoming

in either /etc/MailScanner/MailScanner.conf or a file in
/etc/MailScanner/conf.d
